Advancing Siddha Medicine Delivery Through Bilayer Tablet TechnologyTraditional Siddha medicine includes dosage forms such as Chooranam (herbal powders), Parpam and Chendhuram (herbo-mineral/mineral preparations), which are sometimes administered in combination based on traditional therapeutic principles. However, conventional co-administration may present practical challenges related to dose accuracy, handling of multiple dosage forms, dosing frequency, patient convenience, and compliance.Bilayer tablet technology offers an innovative pharmaceutical approach by incorporating the herbal and herbo-mineral components into a single dosage form with two distinct layers. This approach provides an opportunity to improve dose precision, patient convenience, formulation stability, and flexibility in drug-release characteristics, while retaining the underlying concept of combined administration.Why Bilayer Tablets?🔹 Improved Patient Convenience – Multiple powders or dosage forms can potentially be consolidated into a single tablet, simplifying administration.🔹 Dose Precision – Defined quantities of herbal and herbo-mineral components can support more consistent dosing than manual measurement of separate preparations.🔹 Layer-wise Separation – Physical separation of the components may help minimize undesirable interactions between the two layers prior to administration and supports formulation stability.🔹 Controlled Release Potential – Bilayer design provides flexibility to develop different release profiles, including immediate, sustained, or synchronized release, according to the intended therapeutic objective.Towards Standardization and Scientific ValidationOur review proposes a structured framework for the development, optimization, characterization, and testing of herbal–herbo-mineral bilayer tablets, incorporating modern pharmaceutical approaches such as pre-formulation studies, HPTLC, ICP-OES, FTIR, DSC, XRPD, dissolution and release-kinetics studies, stability assessment, and Quality by Design (QbD).The work aims to provide a scientific foundation for developing standardized and reproducible SOPs for Siddha bilayer tablets, while considering applicable testing frameworks and broader global pharmaceutical development principles.From Traditional Wisdom to Modern Pharmaceutical InnovationThis review represents an effort to bridge traditional Siddha therapeutic concepts with contemporary pharmaceutical technology, with the long-term vision of improving quality, safety, reproducibility, patient acceptability, and scientific validation of Siddha dosage forms.Heartfelt congratulations to the entire research team for this collaborative work, and sincere gratitude to the Central Council for Research in Siddha (CCRS), Ministry of Ayush, for the financial and institutional support provided through the IMR project, “Formulation and optimization of Orally Disintegrating Tablet (ODTs), Single and Bilayer tablet formulations to be developed in Siddha System of Medicine to Enhance Patient Compliance.”Research Team:Dr. M.
